  • Nausea/vomiting, diarrhea
  • Stomatitis
  • Leukopenia, thrombocytopenia
  • Alopecia, photosensitivity, rash

Pharmacology

  • Half-life: 8-15hr
  • Metabolism: Extensive hepatic and intracellular
  • Excretion: Mostly renal. Dialyzable.

Mechanism of Action

  • Inhibits dihydrofolate reductase--> blocks DNA synthesis and thus, cellular replication/repair
